Conversion of 7-OH from Mitragynine

Kratom (Mitragyna speciosa) contains over 40 alkaloids, but two of the most studied are mitragynine and 7-hydroxymitragynine (7-OH). 

🌿Mitragynine is the primary alkaloid in most kratom leaves, accounting for up to 66% of the total alkaloid content.

🌿7-OH, although found in smaller quantities, is known for its significantly stronger affinity to mu-opioid receptors, meaning it is responsible for many of the plant's potent effects.

The Science Behind the Conversion

According to ACS Laboratory7-OH is not typically present in large quantities in raw kratom. Instead, it's formed either naturally through metabolism or synthetically under controlled conditions. 

 

Biological/Natural Conversion

Once mitragynine is ingested, the body's liver enzyme (cytochrome P450 system) begins metabolizing it. During the process:

  • Mitragynine undergoes oxidation, a chemical reaction where it gains oxygen atoms

  • Resulting in the formation of 7-hydroxymitragynine, which can then interact more directly with the body's receptors. 

This conversion is what gives Kratom its unique time-released effects. 

Laboratory-Based Oxidation

Outside the body, scientists can replicate this transformation through oxidation-reduction reactions, using specific chemical agents. Common oxidizers include:

  • Hydrogen peroxide (H2O2)

  • Potassium ferricyanide

  • Other oxygen-donating compounds

Under precise pH and temperature conditions, these agents oxidize mitragynine at the 7-position, resulting in the formation of 7-OH. The process is useful for analytical studies, research, and formulation of kratom extracts.
